I have a (in Spanish) with 800+ videos. So let me tell you how I do the video part I pubsilh all the videos in Youtube and blip.tv using . Tubemogul also allows to pubsilh the videos on an S3 account or to a server via FTP (with conversion of the video to the format that suits you). Extremely recommended.Then I pubsilh the videos on my site using Youtube player and adding the blip.tv files as enclosures, so they are recognised by iTunes and Miro. I also pubsilh it then in and on top of the front page using a WP plugin for Youtube and Vimeo called (I have the paid version, which allows the thumbnails on top) .But if you want something more professional, you might be interested in . Haven't tried it, but looks very good. One day Hope this helps
